The high level of organization exhibited in the cerebellum is revealed in the sagittal rat brain section featured in the image above. Note, nuclei, neural axons, and astrocytes chiefly appear in distinct layers. The cerebellar pattern was visualized by immunofluorescently labeling the specimen with rabbit anti-GFAP antibodies (glial fibrillary acidic protein; astrocytes and certain other stellate cells) and chicken anti-NF-H antibodies (neurofilaments; most abundant in axons of large projections neurons) followed by goat anti-rabbit and anti-chicken antibodies conjugated to Alexa Fluor 568 and Alexa Fluor 488, respectively. Subsequently, cell nuclei were envisaged with the red-absorbing probe DRAQ5 (pseudocolored blue). Images were recorded with a 10x objective using a zoom factor of 1.6 and sequential scanning with the 488-nanometer spectral line of an argon-ion laser, the 543-nanometer line from a green helium-neon laser, and the 633-nanometer line of a red helium-neon laser. During the processing stage, individual image channels were pseudocolored with RGB values corresponding to each of the fluorophore emission spectral profiles unless otherwise noted above.
