FluoView FV1000/IX Tissue Culture Microscope


The Olympus FluoViewTM FV1000 is a next-generation imaging system designed for high-resolution, confocal observation of both fixed and living cells. The FV1000 offers advances in confocal system performance while providing the speed and sensitivity required for live cell imaging with minimal risk of damage to living specimens. In addition, the FV1000 offers a revolutionary synchronized laser scanning system called the SIM Scanner. While one laser stimulates, the second laser simultaneously provides high-resolution imaging. This coordination of laser stimulation and imaging makes the FV1000 an ideal choice for FRAP, FLIP and photoactivation.

Enhanced with specialized applications modules, the FV1000 confocal operations software is designed to operate on standard Windows computer systems with an intuitive, menu-driven interface. Images can be displayed side-by-side in single channels, merged, tiled, and presented with details of the acquisition mechanism. In addition, the software contains look-up tables for individual color settings and pseudo-color, as well as providing the ability to add graphical and text input for comments.

Images collected in a time course experiment or through a serial z series of optical sections can be displayed chronologically as a live tiling display. During scanning, images can be recalled and analyzed at any point. Image acquisition conditions can be recalled with the image files displayed as thumbnails. A simple double click on a thumbnail icon will display that image's acquisition parameters on the interface menu.

Detailed information on the scanning conditions, including each line's scan time, x-y resolution, and field of view within the scan range, can be displayed individually. The area of the specimen and its scanning conditions are also clearly indicated. The operator can easily make adjustments while examining the sensitivity parameters (photomultiplier voltage, gain, offset, etc.). Automatic contrast mode makes image acquisition very easy.

While acquiring xyzt image dimensions and composing three-dimensional images, a separate image of any designated cross section can be obtained. This enables the scanning and image processing conditions to be changed in real time, in effect, while scanning. The operator can observe any region in varying perspective by changing from 2-D to 3-D and vice versa dynamically. In addition, the composed three-dimensional (volume rendered) image can be tilted in any direction to facilitate observation.

During any experiment, sudden and unexpected changes often occur when examining living cells. When they do, many different operating conditions must be changed quickly without interrupting the experimental procedures. The excellent operability and compatibility of the FV1000 software interface enable superb and instantaneous response to such situations, allowing investigations to proceed smoothly.

Many of the FV operating conditions, including the AOTF function, laser exchange, light acceptance channel, and sensitivity setting, can be flexibly altered while scanning. Because it it possible to adjust such setting conditions as interval and synchronization with an external device, the operator can respond quickly to a variety of conditions during time-lapse experiments. In addition, ratio images can be displayed in real time.

Any region of interest (ROI) can be specified by excitation and observation independently, with unrestricted control of variations in timing, duration, and intensity. The FV1000's sophisticated and independent, yet coupled, excitation and scanning systems enable many different scenarios for laser light stimulation.

Unrestricted programming is possible by using a specially developed program language. Experiments systematically programmed with a time series controller can be separated and freely combined, enabling the next experiment to be selected after judging the analysis results of the one before. Comprehensive analysis functions include time series display at any z series in an xyzt image, display of a list of ROI measurement results, and many more features.
