A log phase culture of 3T3 cells was immunofluorescently labeled with primary anti-tubulin mouse monoclonal antibodies followed by goat anti-mouse secondary antibodies conjugated to Alexa Fluor 488. The cells were simultaneously stained for DNA with TO-PRO-3 and imaged with argon-ion and helium-neon lasers. Images were recorded with a 60x oil immersion objective using a zoom factor of 2.0 and sequential scanning with the 488-nanometer spectral line of the argon-ion laser and the 633-nanometer line of the red helium-neon laser. During the processing stage, individual image channels were pseudocolored with RGB values corresponding to each of the fluorophore emission spectral profiles.
